where do the commands go?

I'm really confused, I'm hoping someone can help me understand how this works. When using check_nrpe to check modules on a nscp agent can I run a check directly from the command line of the Nagios server without there being a matching definition in the nsc.ini file under NRPE Handlers (i.e. ./check_nrpe -H hostname -c CheckFileAge? -a .....)? Or do I have to add a command to the nsc.ini file under NRPE Handlers before utilizing any of the modules (other than NSClientlistener) for every check I want to run against an nscp agent?
